Description Purification of Thiobacillusferrooxidans cultures by single colony isolation has been a real problem partly because of the difficulty to grow the cells on a solid medium. As a result many apparently pure cultures of T. ferrooxidans have been found to contain more than one strain or acidophilic heterotrophic bacteria as contaminants. Adoption of an agarose-based solid medium to grow this organism appears dependable either to distinguish the strains according to colony morphology or to purify a strain from heterotrophic contaminants by single colony isolation. That colony morphology of the same strain may differ depending on the source of agarose is another interesting observation which advises one to maintain strictly identical growth conditions to distinguish strains according to colony morphology.
