<span style="mso-bidi-language:HI">3,5-Bis(arylmethylene)-4-piperidone derivatives have emerged as novel clusters of cytotoxic agents. The focus <span style="mso-bidi-font-family:Arial;mso-bidi-language:HI">of <span style="mso-bidi-language:HI">the current research has been to tine <span style="mso-bidi-font-family:Arial;mso-bidi-language:HI">tune <span style="mso-bidi-language:HI">the biological activity of these compounds by rational alteration of <span style="mso-bidi-font-family:Arial; mso-bidi-language:HI">substituents <span style="mso-bidi-language:HI">on this <span style="mso-bidi-language:HI">pharmacophore to <span style="mso-bidi-font-family: Arial;mso-bidi-language:HI">increase potency <span style="mso-bidi-language: HI">and selectivity. These compounds arc presumed to bc devoid of <span style="mso-bidi-font-family:Arial;mso-bidi-language:HI">gcnctoxicity <span style="mso-bidi-language:HI">and act <span style="mso-bidi-font-family: Arial;mso-bidi-language:HI">by <span style="mso-bidi-language:HI">selective thiolation of proteins. Anticancer behavior of nine clusters of 3,5-bis(arylmethylene)-4-piperidone derivatives have <span style="mso-bidi-language:HI">been reviewed here with emphasis on their chemistry, cytoxicity and quantitative structure-activity relationship studies. </span></span></span></span></span></span></span></span></span></span></span></span></span></span></span>
