Conventional ion-sensitive sensors, such as glass membrane electrodes, coated wire electrodes, metal-metal oxide electrodes, etc, convert a non-electrical quantity into an electrical quantity, which can then be measured with an appropriate electronic circuitry connected in series. Due to loading effect, there are unpredictable deviations in the transducer parameters that cannot be controlled in such a measuring system. Because the ISFET also has an electrical input besides the ion-sensitive input, this results in unique application possibilities, with the consequences for the development of adequate electronic circuitry . In order to obtain complete knowledge of ISFET as a biosensor, It is useful first to understand the electrical behavior of a MOSFET, from which the ISFET behavior can be derived. Based on this theory some modifications have been made in the physical structure of MOSFET which serves as an appropriate ISFET biosensor capable of measuring pH, urea, calcium and other ions activity in the solution. The paper describes the operation of ISFET based on MOSFET theory and discusses the elimination of problems in the conventional ion-sensitive sensors.
