This study used shake flask method to examine effects of pH, temperature, and duration of incubation on removal efficiency by studying physicochemical parameters and floc stability in degradation of pulp and paper mill effluent over 24 h using mixed cultures of bacteria and fungi. A 2<sup>4</sup> full factorial central composite design and response surface methodology were adopted to plan experiments and analyze the data, respectively. Quadratic models were developed based on high coefficient of determination (R<sup>2</sup>) obtained from analysis of variance (ANOVA) results. Optimal conditions yielded a decrease in chemical oxygen demand (COD,70%) and in total suspended solid (TSS, 53%) levels.
