A rational approach has been adopted for the synthesis of novel series of 3-((6-(2-chloro-6-methoxyquinolin-3-yl)-4-aryl-1,6-dihydropyrimidin-2-yl)thio)propanenitriles <strong>5a-o</strong>. The synthesized compounds have been evaluated for their <em>in vitro </em>antimicrobial activity. Compounds <strong>5j-m</strong> display broad spectrum antibacterial activity against all the bacterial strains. Moreover, compound <strong>5m</strong> is found to be the most potent antibacterial and antifungal agent. Further, the results of preliminary MTT cytotoxicity studies on HeLa cells suggest that potent antimicrobial activity of <strong>5j-m</strong> is accompanied by low level of cytotoxicity.
