Novel pyrazoline derivatives incorporating chalcone backbone have been synthesized, characterized and evaluated for their anti-inflammatory activity. The structural features of synthesized compounds have been established using IR, <sup>1</sup>H NMR, mass spectroscopy and elemental analysis. <em>In vivo</em> anti-inflammatory activity has been measured using carrageenan induced paw edema method. Molecular docking study of synthesized compounds has also been performed using cyclooxygenase-2 (COX-II) receptor PDB ID: 4-COX. Some of the synthesized compound show remarkable anti-inflammatory potential. The compound (<em>E</em>)-1-(5-(4-chlorophenyl)-3-(4-hydroxyphenyl)-4,5-dihydropyrazol-1-yl)-3 (4methoxyphenyl)prop-2-en-1-one <strong>6h</strong> shows potent anti-inflammatory activity. The outcome of docking studies also support results of <em>in vivo</em> anti-inflammatory activity.
