Enhanced production of polyunsaturated fatty acid docosahexaenoic acid by thraustochytrid protists

Description The polyunsaturated fatty acid docosahexaenoic acid (DHA) is an important requirement in the human diet. It is also essential in the nutrition of crustaceans and aquaculture animals. Of the sources available for commercial production of DHA, thraustochytrids are considered among the most promising. These marine eukaryotic, straminopilan protests have been extensively studied in recent years for DHA production. This paper examines methods to enhance DHA production in thraustochytrids. A cold shock treatment during growth of several thraustochytrids resulted in pronounced increases in DHA. An alternative method of harvesting the cells and storing the cell biomass at a low temperature also elevated DHA production. Storage at 10 degrees C for 24 to 48 hours produced the best results. Increase of viscosity of the culture medium by addition of polyvinyl pyrrolidone also enhanced DHA production. Different mechanisms of DHA increase were observed in the present study. In some isolates, DHA seemed to increase proportionately to increased levels of lipid production. In orders, the percentage of DHA contents in lipids increased, as well as the levels of lipid production. Some of the isolates did not exhibit any DHA increase under the experimental conditions. The isolates studied differed in their mechanisms of DHA production. This study suggests that isolates producing moderate levels of DHA can be treated further to produce high levels of DHA.
