Description The synthesis of conformationally flexible medium sized ring ethers, constituting the unique structural core of a large number of linearly condensed cyclic polyether marine neurotoxins, has received considerable attention. Although there are numerous methods for the generation of 7-, 8-, and 9- membered chiral ring ethers, elaboration of the ring poses considerable problem in organic synthesis. Recent developments in establishing generalized synthetic methodologies for seven to nine membered chiral ring ethers are mainly based upon intramolecular cyclization processes, namely (i) ring closing metathesis (RCM), (ii) palladium-induced reaction (Heck reaction), and (iii) radical cyclization. In the last two decades intramolecular radical and Heck reactions have emerged with enhanced importance for the synthesis of natural products, including those incorporating medium sized ring ether systems. During the synthesis of such systems found to benzo- and pyrido- rings, the investigation of steric and electronic effects of the substituents in the ring on the yield and regioselectivity of the product became very important.
