Description Gastric carcinoma is the fourth most cause of cancer related death burdening our healthcare system (Ferlay et al., 2010). The pathogenesis of gastric cancer involves a number of events, including exposure to chemical carcinogens, H. pylori infection and stress (Fox and Wang, 2007). Individual host immunity and genetic polymorphisms are major factors influencing degree of tumor invasiveness and poor patient survival in variety of cancers and thus warrant for exploring the relationship between genetic variations of MMP genes and gastric cancer occurrence and progression. Population of India is extremely diverse. In this regard it is highly expected that genetic screening would result in determination of SNPs of MMP genes associated with gastric carcinoma in an eastern Indian population. The aim of the current investigation is to determine the association of MMP gene promoter SNPs existing in a large gastric cancer patient vs. control study population in Eastern Indian Region. The objectives of the present studies are as follows: 1. Investigate the demographic and clinicopathological characteristics of gastric cancer patients and analysis of MMP gene promoter polymorphisms in eastern Indian population. 2. Investigate the association between polymorphisms in MMP gene promoters and the development and progression of gastric cancer with respect to the clinicopathological features of gastric cancer. Determination of the functional significance of these SNPs by analyzing their promoter activity in silico. 3. The haplotype and diplotype based approach is to be applied to investigate the entire promoter region across MMP1-MMP3-MMP7-MMP9-MMP12-MMP13 genes and to identify single marker and their combinations contributing to the risk for gastric cancer with respect to their LD structure in eastern Indian population. This study may also lead to generate a list of functional SNPs associated with predisposition and progression of gastric cancer for Eastern Indian population which can enrich the population based national genomics database for future personalized cancer therapeutics and possible target for efficient management of gastroduodenal cancer in country like India with vast genetic diversity.
