Description Nano-liposomes were designed by using Supercritical (SC) Gas Anti-solvent (GAS) method. The present approach configured to combine two steps i,e 1) SC−CO2 assisted drying of phospholipids to form uniform layer; 2) hydration under pressure and depressurization of SC liquid phase to generate nanoliposomes. Effect of pressure, temperature and surfactant concentration (Tween 80) was evaluated for production of nano-liposomes. SC−GAS method was compared with Bangham’s method in terms of mean diameter, coefficient of uniformity, morphology, stability index and actual energy required for liposomes formation. Liposomes obtained by Bangham’s method were multi-lamellar vesicles having coefficient of uniformity (12.07 ± 0.024) with mean diameter of 4.64 ± 0.010 μm, exhibiting irregular thickness of bilayer. Liposomes production by SC−GAS method at optimized formulation conditions i,e 170 bar, 50 °C, 1.5% Tween 80 and depressurization rate of 25 bar/min yielded nano-liposomes exhibiting coefficient of uniformity (1.10 ± 0.012) with mean diameter of 0.063 ± 0.002 μm. Stability index and viscosity of prepared liposomes suspension have indicated greater stability and near Newtonian behaviour respectively. SC−GAS process requires 1.52 times lesser energy compared to Bangham’s method to achieve nano-scale liposomes production. Storage studies indicated that SC−GAS based nano-liposomes were highly stable up to 3 months at different temperatures.
